ABSTRACT

Objective To investigate the anti-inflammatory effect of chlorogenic acid( CGA) on BV2 microglia in-fected by herpes simplex virus ( HSV)-1.Methods BV2 cell was stimulated by HSV-1 to create a cell model of vi-rus encephalitis, then CGA was processed with different concentrations. MTT method was used to measure cell via-bility. Real-time quantitative PCR and ELISA were used to detect the content and expression of tumor necrosis fac-tor alpha ( TNF-alpha) and interleukin ( IL)-6. Results After CGA treatment, the cell survival rate significantly improved, and after HSV-1 infected BV2 cells, CGA inhibited the significant increase in mRNA levels of TNF-al-pha and IL-6. In addition, CGA can reduce the release of TNF-alpha and IL-6 induced by HSV in the cells.The mRNAs of TNF-alpha and IL-6 were also significantly inhibited by CGA.Conclusions CGA inhibits inflammatory response in HSE.CGA can be used as an anti-inflammatory agent to treat HSE new strategies.

ABSTRACT

Objective To evaluate the safety and efficacy of recombinant bovine basic fibroblast growth factor in the treatment of corticosteroid-dependent dermatitis.Methods A randomized,double-blind,placebo-controlled clinical trial was carried out.By simple randomization,64 patients with corticosteroid-dependent dermatitis were randomly and equally divided into 2 groups:treatment group topically applying recombinant bovine basic fibroblast growth factor gel twice a day for 4 consecutive weeks,and control group topically applying the gel vehicle twice a day for 4 consecutive weeks.Clinical symptoms and signs were scored before the treatment and after 1-,2-and 4-week treatment.Meanwhile,the water content of the stratum corneum,skin sebum content and transepidermal water loss (TEWL) of the skin lesions were detected.Results Thirty-one patients in the treatment group and 30 in the control group completed the trial.The clinical symptom and sign scores in the treatment group were significantly lower at week 2 and 4 after starting treatment (1.35 ± 0.55 and 1.00 ± 0.45,respectively) than that before treatment (2.77 ± 0.43,both P < 0.05),as well as lower at week 1 (2.06 ± 0.51),2 and 4 after starting treatment than that in the control group (2.43 ± 0.57,2.17 ± 0.53,1.93 ± 0.45,respectively,all P < 0.05).The treatment group showed significantly increased water content of the stratum corneum at week 4 after starting treatment,significantly increased skin sebum content,but decreased TEWL at week 2 and 4 after starting treatment compared with those before treatment (all P < 0.05).Compared with the control group,the treatment group showed significantly higher skin sebum content at week 2 and 4 after starting treatment,higher water content of the stratum corneum,but lower TEWL at week 4 after starting treatment (all P < 0.05).No adverse reactions were observed in either of the 2 groups.Conclusion Recombinant bovine basic fibroblast growth factor is effective and safe for the treatment of corticosteroid-dependent dermatitis,and contributes to repairing and reconstructing the skin barrier function.

ABSTRACT

Objective To explore the effects of Sishen Pills on Ghrelin-CaM-MLCK signaling pathway with sinuses ventriculi in diarrhea rats of yang deficieney of spleen and kidney; To discuss its mechanism of action. Methods A model rats with diarrhea model of yang deficieney of spleen and kidney was established by treated with intragastric administration of adenine and Senna water decoction to replicate. 72 SD rats were randomly divided into normal group, model group, positive medicine group, Sishen Pills high-, medium- and low-dose groups, with 12 rats in each group. Each medication group was given relevant medicine for gavage, for 14 days. Immunohistochemistry and Western blot were used to detect the expressions of Ghrelin and GHSR, CaM, and MLCK protein in the sinuses ventriculi. Results In the model group, Ghrelin, GHSR, CaM and MLCK protein expressions of the sinuses ventriculi significantly increased (P<0.01). In each medication group, protein expression of the indexes was lower than the model group (P<0.05, P<0.01). The Sishen Pills high-dose group was particularly obvious. Conclusion The effect of Sishen Pills on diarrhea model rats of yang deficieney of spleen and kidney may be associated with regulation of index protein expression in Ghrelin-CaM-MLCK signaling pathway.

ABSTRACT

Objective To investigate the effects of immune system and immuno-related miRNAs expression in low-dose benzene and its homologue mixed-exposed workers in a short time. Methods A total of 40 workers were recruited from a petrochemical factory ,and their blood samples were collected before and after work to detect the levels of IgA,IgG,IgM,IFN-γ,IL-1β,IL-2,IL-8,TNF-αin serum and miRNA-146a,-155 expression in the peripheral blood mononuclear cell. In the meanwhile ,we measured the individual benzene and its homologue exposure level of recruited workers. Results According to the concentration of benzene ,the subjects were divided into benzene-exposed group(20 workers)and control group(20 workers). And toluene exposure level in benzene-exposed group was significantly higher than that in control group(P<0.05). Significant decrease of IgG was found in benzene-exposed workers after one work shift,compared with control group(P<0.05). Moreover,the interaction between benzene and toluene was significant correlated with the decrease of IL-1β and TNF-α(P < 0.05). Additionally,the interaction between miRNA-155 and miRNA-146a was significant correlated with the decrease of TNF-α(P<0.05). Conclusions:Occupational exposure to low-dose benzene and toluene in a short time could decrease immune function. And there may be an effect of miRNAs in the regulation of cytokine.

ABSTRACT

AIM:To investigate the effect of angiotensin II type 1 receptor (AT1R)-calcineurin (CaN) signa-ling pathway on the expression of sodium current channel Nav 1.5 at mRNA and protein levels in the hypertrophic ventricu-lar myocytes from neonatal rats .METHODS:The ventricular myocytes were isolated from the ventricles of 1-day-old neo-natal Sprague-Dawley rats and were divided into 4 groups according to different drug intervention as control group , pheny-lephrine (PE) group, losartan (Los)+PE group and cyclosporin A (CsA)+PE group.The method of RNA interference mediated by adenovirus carrying short hairpin RNA ( shRNA) was used to knock down the gene which encodes the beta subtype of CaN A subunit (CnAβ) and the cells were divided into 4 groups as Ad-Null group, Ad-Null+PE group, Ad-CnAβshRNA1 group and Ad-CnAβshRNA1+PE group.The mRNA expression of brain natriuretic peptide ( BNP) ,β-my-osin heavy chain (β-MHC) and Nav1.5 was detected by RT-qPCR.The protein levels of CnAβand Nav1.5 in the whole-cell extracts were determined by Western blot analysis .RESULTS:Treatment of the neonatal rat ventricular myocytes with PE for 24 h increased the protein-to-DNA ratio and the mRNA expression of BNP and β-MHC.The size of the cell surface was also increased after PE treatment .Treatment of the cells with PE increased the protein expression of CnAβ, and re-duced the protein expression of Nav 1.5.Both Los and CsA prevented those effects of PE .The mRNA expression of Nav1.5 was reduced by PE , and no significant difference of Nav 1.5 mRNA expression among PE group , Los+PE group and CsA+PE group was observed .Silencing of CnAβin the neonatal rat ventricular myocytes using Ad-CnAβshRNA1 inhibited the ability of PE to increase the mRNA expression of BNP , and diminished the ability of PE to reduce the protein expression of Nav1.5.CONCLUSION:AT1 R-CaN signaling pathway participates in regulating protein expression of Nav 1.5 in the hy-pertrophic ventricular myocytes from neonatal rats .

ABSTRACT

AIM:To study the influence of lithium chloride (LiCl) on the neuronal differentiation of rat bone marrow mesenchymal stem cells (MSCs), and to explore whether autophagy was involved in this process .METHODS:MSCs were isolated and cultured in vitro.The cells were divided into LiCl group and control group .MSCs were treated withβ-mercaptoethanol as an inducer for triggering the cells to differentiate into neurons .The expression of neuronal markers-neuron specific enolase (NSE) and microtubule-associated protein-2 (MAP-2), and autophagic marker-microtubule-associ-ated protein 1 light chain 3 ( LC3) were measured by immunofluorescence method and Western blot .An autophagy activator rapamycin and autophagy inhibitor 3-methyladenine (3-MA) were applied to modulate the autophagy in the LiCl treated-cells.The protein expression of NSE and MAP-2 were determined by Western blot .RESULTS: After induction, the ex-pression of NSE and MAP-2 were increased .The percentage of NSE-and MAP-2-positive cells and the expression of NSE and MAP-2 in the LiCl group were greater than those in control group (P<0.05).After induction, the number of LC3-positive dots and the expression of LC3-Ⅱin LiCl group were greater than those in control group (P<0.05).The expres-sion of NSE and MAP-2 increased when the autophagy was modulated by rapamycin in LiCl treated -cells, and on the contra-ry, the expression of NSE and MAP-2 were inhibited as autophagy was modulated by 3-MA.CONCLUSION: Lithium chloride may promote the neuronal differentiation of rat bone marrow mesenchymal stem cells by modulating autophagy .

ABSTRACT

A new napthalenone, rumexone A (1), was isolated from the roots of Rumex nepalensis. The structure of 1 was elucidated by extensive spectroscopic analyses, including 1D and 2D NMR spectra and MS data. Its cytotoxic effect was evaluated using four clinically relevant human cancer cell lines, gastric carcinoma SGC7901, breast carcinoma MDA-MB-231, lung carcinoma A549, and hepatocellular carcinoma HepG2.

ABSTRACT

Objective: To explore the role of angiotensin receptor type I (AT1)-calcineurin (CaN) signaling pathway in transient outward potassium ion channel (Ito) remolding in hypertrophic atrial myocytes of neonatal rats. Methods: 1 day old neonatal rats’ atrial myocytes were isolated and the cells were divided into 4 groups:①Control group, normal cells were cultured for 24 h,②Stretching group, the cells were cultured for 24 h with mechanical stretching to induce hypertrophy,③Telmisartan group, the cells were treated by telmisartan at 1 μmol/L for 1 h, then cultured for 24 h and ④Cyclosporin-A (CsA) group, the cells were treated by CsA at 0.25 μg/ml for 1 h, then cultured for 24 h. The ratios of protein/DNA in myocytes were compared between Control group and Stretching group, cell hypertrophy was deifned by mRNA expression of atrial natriuretic peptide (ANP). Ito changes were detected by whole-cell patch clamping technique, proteins expressions of Kv4.3 and CaN A subunit were examined by Western blot analysis. Results: Compared with Control group, Stretching group showed obviously decreased Ito density and Kv4.3 protein expression, while increased CaN A protein expression; Compared with Stretching group, the above effects were reduced in Telmisartan group and CsA group. Conclusion: AT1-CaN signaling pathway was involved in the regulation of Ito channel remodeling in hypertrophic atrial myocytes of neonatal rats.

ABSTRACT

Objective: To investigate the chemical constituents from the ethyl acetate extracts in the roots of Rumex nepalensis. Methods: The compounds were isolated by using various chromatographic techniques, such as silica gel column, Sephadex LH-20 column, and semi-preparative HPLC. The structures of these compounds were determined by spectral data analyses. Results: Sixteen compounds were obtained and identified as ferulic acid (1), 7-hydroxy-5-methoxyphthalide (2), methyl 2-acetyl-3,5-dihydroxyphenylacetate (3), methylorsellinate (4), methyl p-hydroxycinnamate (5), 1-(2-Hydroxy-5-Methyl-phenyl)-ethanon (6), methyl syringate (7), 1-(2,4-dihydroxy-6-methylphenyl)-ethanon (8), 4-hydroxy-benzeneethanol (9), isovanillin (10), fallacinol (11), 7-hydroxy-2-methyl-5-methylchromone (12), 3-acetyl-2-methyl-1,5-dihydroxy-2,3-epoxynaphthoquinol (13), emodin (14), chrysophanol (15), and physcion (16). Conclusion: Compounds 12 and 13 are isolated from this plant for the first time, and compounds 1-11 are firstly isolated from the plants of Rumex L..

ABSTRACT

<p><b>OBJECTIVE</b>This study aims to evaluate the quality of life (QOL) of patients who underwent resection of oral cancer and reconstruction by free anierolateral thigh perforator flaps (ALTF).</p><p><b>METHODS</b>A total of 32 patients with oral and maxillofacial malignancies who had undergone the resection of oral cancer and reconstruction by ALTF were retrospectively analyzed. At 12 months postoperatively, the QOL of these patients was assessed by using the 14-item oral health impact profile (OHIP-14) and the medical outcome study short form-36 (SF-36) questionnaires.</p><p><b>RESULTS</b>A total of 32 questionnaires were collected. In SF-36, the highest scoring domains were bodily pain (78.58 ± 14.82), physical functioning (72.08 ± 27.86), and the role of physical (60.00 ± 42.63), whereas the lowest scoring domains were role-emotional (41.67 ± 39.62), followed by mental health (50.75 ± 13.07) and health transition (54.17 ± 21.75). In OHIP-14, the lowest scoring domains were social disability (34.50 ± 11.32) and handicap (36.04 ± 12.05), indicating the functional recovery was better; and the highest scoring domains were physical pain (73.50 ± 18.96) and psychological discomfort (60.17 ± 20.66), indicating the functional recovery was worse.</p><p><b>CONCLUSION</b>The ALTF is an ideal selection for the reconstruction of oral defects after cancer resection. In using this flap, the basic social need of patients after surgery can be satisfied. Moreover, the appearance and the functions of chewing, deglutition, and speech can be restored in varying degrees. Thus, ALTF can improve the patients' QOL.</p>

ABSTRACT

Objective To investigate the expression of intestinal epithelial stem cell markers Msil and Hesl in mouse acute radiation enteritis (RE) model. Methods Forty female BALB/c mice were undergone one-time whole abdominal irradiation by 300 cGy/min β-ray. Thirty mice were selected before irradiation (Day 0) and every six mice were euthanized at day 3,5,7 and 14 after irradiation. The small intestinal were detached and the expression of Msil and Hesl were detected in middle part of small intestinal by quantitative RT-PCR, Western blots, and immunohistochemistry. Subsequently,the difference of Msil and Hesl expression was compared. Results The mouse acute RE model was successfully established. At the 5th day after radiation, the results of quantitative RT-PCR indicated that the relative ΔCt value of Msil and Hesl expression were 15. 17 ± 0. 47 and 15. 83 ± 0. 24respectively, the results of Western blot showed that the integrated intensity were 0. 443±0. 055 and 0. 301 + 0. 047 for Msil and Hesl. The expressions were significantly higher than other time points at RNA and protein levels (P<0. 05). The results of immunohistochemistry revealed that Msil and Hesl were highly expressed in intestinal crypts at 5th day and 7th day. Conclusions The expression of Msil and Hesl, upregulated before wound repair in RE model, indicates that intestinal epithelial repair is related to intestinal stem cell proliferation.

ABSTRACT

Objective. To observe the effect of chemotherapy on hepatic function of lymphoma patients with chronic HBV infection. Methods; We used ELISA to detect the serum markers, of HBV and liver function in 207 lymphoma patients and 207 patients with other types of cancer (except pdmary hepatocellular cacinoma). Results: The incidence of HBV infection was higher in lymphoma cancer cases than that in the controlled cases (19.8% vs 9.7%, P=0.004). The incidence of abnormal liver function was higher in lymphoma patients with positive HBsAg than in lymphoma patients without HBsAg (58.5% vs 27.7%, P=0.000). The incidence of ab-normal liver function in lymphoma patients with postive HBsAg was higher than that in patients with other types of cancer with positive HBsAg (58.5 vs 30.0%, P=0.036). The abnormal liver function in lymphoma patients after chemotherapy was associated with HBV infection (P=0.000) but not correlated with age, sex, histological subtype, immune subtype, stage, ECOG PS, and hormone administration. Conclusion: Lymphoma patients with HBV are more likely to have liver function damage after emotherapy.

ABSTRACT

Objective To introduce the gender-specific view to the prenatal care and analysing the health behavior and its influencing factors. Methods Random sampling in cluster was used to select those study population and face to face interview was carried out at their residence in Xinjiang and Anhui province. Results The overall socioeconomic status of women in rural areas of Xinjiang and Anbui province was low and 93.2% of the women had education level on junior middle school or below. Politically women were less interested in these issues with only 63.1% of them ever participated in the election program or voting. In household daily life, women' s status was almost equal to or even higher than men, but men always called the shots when something important in family happened. Men still held the power of decision making. In 47.3% of the families, the husband played a decisive role on decision making. In 37.7% of the families, husbands controlled the money. When getting into expensive expenditure, 35.2% of the families having men made the final approval. When there were different opinions between the couples, husband usually made the final decision in 44.2% of all the families compared to 6.3% of the families that wives made the final decision. 73.9% of women under survey bad undergone prenatal examination and the rate on household deliveries reached 30.3%. Both gender consciousness and educational level had influenced the effect on prenatal care. Conclusion It is important to better prenatal health service in rural areas and to improve socioeconomic status on gender consciousness of women.

ABSTRACT

<p><b>OBJECTIVE</b>To study the application of decision tree in the research of anemia among rural children.</p><p><b>METHODS</b>In the Enterprise Miner module of software SAS 8.2, 3000 observations were sampled from database and the decision tree model was built. The model using decision tree of CART bases on Gini impurity index.</p><p><b>RESULTS</b>The misclassification rate of decision tree model was, training set 21.2%, validation set 21.9%. The Root ASE of decision tree model was, training set 0.399, validation set 0.404. The area under the ROC curve was larger than the reference line. The diagnostic chart showed that the corresponding percentage was higher than the other. The decision tree model selected 9 important factors and ranked them by their power, among which mother of anemia (1.00) was the most important factor. Others were children's age (0.75), time of ablactation (0.53), mother's age (0.32), the time of egg supplementation (0.26), category of the project county (0.26), the time of milk supplementation (0.16), number of people in the family (0.13), the education status of the mother (0.12). Decision tree produced simple and easy rules that might be used to classify and predict in the same research.</p><p><b>CONCLUSION</b>Decision tree could screen out the important factors of anemia and identify the cutting-points for factors. With the wide application of decision tree, it would exhibit important application values in the research of the rural children health care.</p>

ABSTRACT

Age-related macular in human retinal results in strict loss and enduring decrease in visual acuity. A lot of medical evidence has shown that macular degeneration, associated with Choroidal neovascularization (CNV), leads to irreversible injure in composition and function of retinal tissue. A new analytical and restorative treatment, scotoma-based photocoagulation (SPB), curdles the affected areas so effectively and timely that it can delay or prevent age-related macular degeneration (AMD). A scanning laser ophthalmoscope (SLO) image is involved and the function and character of scotometry are analyzed. Combining anatomical and pathological features, this paper introduces process of acquiring the image and algorithms of analyzing it. Furthermore, integrate image of retinal is extracted from SLO through feature extraction, special registration and superimposition. The treatment can be applied to clinical diagnosis without great revisions and provide direction for pertinent fields.
